Project Description
Fungal diseases are largely responsible for grapevine decline causing extensive losses in production yields. This presents a major problem for the vine and wine industry. Eutypa dieback (Eutypa lata / Eutypa armeniacae) and black dead arm (Botryosphaeria spp.) are responsible for infections of asymptomatic nursery material. The grapevine trunk diseases caused by Eutypa lata and the Botryosphaeria spp. have become an important area of study because of their devastating impact on vineyards worldwide. These canker diseases may limit the long term sustainability of wine and grape production by limiting the productivity of new vines and can be costly because of the expense of vine destruction, replanting, and importantly, the magnitude of yield losses that can occur.
Because the diseases appear to be latent, it becomes vital for farmers to know that cuttings are healthy before planting them in vineyards. Subsequently it is important to develop an identification method of these diseases even on young nursery material that are asymptomatic. The development of a molecular technique for screening nursery material and isolates would be important not only in fungicide screening but in screening for resistant rootstocks too (it is known that certain cultivars show varying levels of susceptibility to the diseases). With a view to developing a screening technique that would provide fast, accurate and reliable means of detecting plant pathogens, reverse dot blot technology appears promising.
